焦点是搞清楚精子如何通过引起细胞内钙离子浓度瞬间升高(一个到目前为止在所研究的动物种类中均观察到的现象),启动储存在卵子内的胚胎发育程序。这个钙信号过程在卵激活和早期胚胎发育中起关键作用。我们的研究证据显示在哺乳动物卵子内钙振荡的有序进行主要依赖于两个必要因子:一个是触发钙从细胞内钙库释放的来自精子内的蛋白因子;而另一个是决定振荡过程是否可以持续的母源性装置。我们已证明来源于精子的蛋白因子的活性在进化上是高度保守的;从植物精子中提取的提取物显微注射到小鼠卵内可以诱导小鼠卵发生钙振荡。我们正准备纯化这个精子蛋白因子并搞清楚这个因子如何通过IP3受体使钙从细胞内钙库中释放的。
我们首次报道了精子因子诱导的钙振荡是受哺乳动物卵子内一个母源性装置调控的,该装置的功能是一次性的。现在我们要研究的是这个母源性装置的性质和特性。此外,我们要确定精子蛋白因子用于触发钙从细胞内钙库释放的特异信号传导通路。
